Obesity and type 2 diabetes (T2D) are major risk factors for cardiovascular (CV) diseases. Dysregulated pro-apoptotic ceramide synthesis reduces β-cell insulin secretion promotes hyperglycaemic states that may manifest as T2D. Sirtuin-1 (SIRT1) is a NAD + - dependent deacetylase that protects against pancreatic β-cell dysfunction. A new study by Dr. Srividya Velagapudi and colleagues shows how SIRT1 improves glucose metabolism in type 2 by inhibiting de novo ceramide synthesis.
